So I started installing weather strip and when I got to the roof rails I notice that I did not have the hole for the screw that holds the strip in place at the bottom of the windshield pillar wear the molding ends. Hole is missing on both sides so is it possible the very early cars were just glued in?

Yes, that's right, the tab wasn't seen until much later in '65 production.
